Mizruchi's father [[Ephraim Mizruchi|Ephraim]] was a sociologist.<ref name="eobit"></ref> Her mother Ruth was of Israeli Jewish descent.<ref name="robit"></ref> [[Mark Mizruchi]] is her elder brother.<ref name="eobit"/><ref name="robit"/> Susan Mizruchi attended [[Washington University]] graduating with bachelor of arts degrees in English and history in 1981.
